The 22-year-old's black Mercedes-Benz was involved in a collision with a truck on a main road not far from the club's training ground in Sinsheim.
He was airlifted to a hospital after receiving life-threatening head injuries, according to a statement from the Bundesliga club.
Vukcevic underwent surgery immediately upon arrival at the clinic and is reported to be in a "critical condition" with no prognosis for recovery.
Vukcevic has been a regular in the Hoffenheim side since making his debut in May 2009, playing a total of 78 times, and he also has been capped by Germany's under-21s.
"This news has shaken us," said Hoffenheim's general manager Andreas Muller. "The team, the coach and everybody here are shocked. Our thoughts are with Boris and his family."
Hoffenheim's Bundesliga clash with Augsburg went ahead Saturday afternoon and ended in a scoreless draw.
"This is the most difficult situation the team has ever been in," the club's general manager Andreas Muller said at a press conference before the match. "We would have been able to call the game off, but the team decided unanimously to play. We want our positive energy to be transmitted to the lad. We want to fight for Boris because he is also a fighter."
Their decision to take to the field at the Rhein-Neckar Arena this afternoon was taken after receiving a message from the player's parents.
"After receiving that, the team and the club decided to play," Muller added.
Neither side managed to create any major chances to win the points as thoughts were clearly elsewhere Saturday.
Hoffenheim's players chose not to request a postponement of the game with their team-mate lying in intensive care.Instead, they emerged for the prematch warm-up with shirts dedicating their efforts to the Croatian midfielder.

I wish the new news was better but at least he is stable now. Giving the players two days off was absolutely the right move here. I'm actually really happy about this, mainly for the fact that I for some reason expected bad news with such a late bump.

Wow that news is awesome, after looking at all the pictures I quite honestly did not expect to here this. Great to see this kid is actually expected to make a full recovery.
